Aceclofenac API: NSAID Uses, Formulations, and Safety Highlights
Technical and clinical overview of Aceclofenac API, a widely used NSAID for pain and inflammation, including its mechanism, indications, and formulation considerations.
What is Aceclofenac API?
Aceclofenac is a non‑steroidal anti‑inflammatory drug (NSAID) structurally related to diclofenac. It is commonly used for the treatment of painful inflammatory conditions such as osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis. As an API, Aceclofenac is supplied mainly for oral solid dosage forms.
Therapeutic Indications
- Osteoarthritis – relief of joint pain and inflammation
- Rheumatoid arthritis – control of chronic inflammatory symptoms
- Ankylosing spondylitis – management of pain and stiffness
- Other musculoskeletal conditions where NSAID therapy is appropriate
Mechanism of Action
Aceclofenac primarily works by inhibiting cyclo‑oxygenase (COX‑2 > COX‑1) enzymes, reducing the synthesis of prostaglandins that mediate pain, inflammation, and fever. Compared with some older NSAIDs, it may have slightly better gastrointestinal tolerability, though GI risk still exists.
Common Finished Dosage Forms
- Film‑coated tablets, typically 100 mg strength (once or twice daily)
- Combination tablets with paracetamol, serratiopeptidase, or other agents (depending on market)
Quality and Stability
Quality Aceclofenac API should exhibit controlled levels of related substances, low residual solvents, and consistent particle size for robust dissolution. Stability studies should cover sensitivity to moisture and light, with packaging designed to minimise degradation.
Safety Considerations
As with all NSAIDs, finished Aceclofenac products can cause gastrointestinal irritation, ulceration, renal effects, and cardiovascular risks in susceptible patients. They should be used at the lowest effective dose for the shortest duration. At the API level, standard PPE, dust control and safe handling practices should be followed.
